Perinatal Changes in leptin receptors expression in liver of rat

الملخص

Leptin is adeponectin that plays a role in organogenesis of different organs through its receptors (OBR), its role in liver organogenesis is not clarified yet; therefore this study aimed to evaluate the immunohistochemical expression of leptin receptors in liver parenchymal tissue during perinatal development and it's relation to hepatocytes maturation and glycogen deposition. Ten adult female rats (Rattus rattus norvegicus) were kept for fertilization with 5 male rats in plastic cages; embryos of them were divided into: Group A embryos at 14 days, Group B embryos at 18 days gestation respectively (prenatal period), (postnatal period): Group C animals at 7 days and Group D animal control adult female non pregnant rats. The liver tissues were fixed in 10% NBF and embedded in paraffin wax, sections were stained with haematoxylin and eosin, PAS stain used to evaluate glycogen in hepatocytes as maturation index and anti leptin-receptors mouse monoclonal antibody Ob-R(B3) SC-8391. That evaluated by Aperio Scope Image analysis software. Result showed significant increase in immunohistochemical expression of leptin receptor in liver tissue at group A that reduced significantly in group B & group C, while glycogen not appearin group A but appeared at the later stages at group B and C, this proof a role of leptin in hepatocytes maturation and may involve in their early sensitization to insulin that lead to glycogen deposition at later stages. © 2020, Connect Journal.
